Caffeine-induced impairment of glucose tolerance is abolished by beta-adrenergic receptor blockade in humans.

چکیده
The caffeine-induced impairment of insulin action is commonly attributed to adenosine receptor (AR) antagonism in skeletal muscle. However, epinephrine, a potent inhibitor of insulin actions, is increased after caffeine ingestion. We tested the hypothesis that the insulin antagonistic effects of caffeine are mediated by epinephrine, and not by AR antagonism, in seven healthy men. On four separate occasions, they received 1) dextrose (placebo, PL), 2) 5 mg/kg caffeine (CAF), 3) 80 mg of propranolol (PR), and 4) 5 mg/kg caffeine + 80 mg of propranolol (CAF + PR) before an oral glucose tolerance test (OGTT). Blood glucose was similar among trials before and during the OGTT. Plasma epinephrine was elevated (P < 0.05) in CAF and CAF + PR. Areas under the insulin and C-peptide curves were 42 and 39% greater (P < 0.05), respectively, in CAF than in PL, PR, and CAF + PR. In the presence of propranolol (CAF + PR), these responses were similar to PL and PR. These data suggest that the insulin antagonistic effects of caffeine in vivo are mediated by elevated epinephrine rather than by peripheral AR antagonism.

Role of adrenergic receptors in the caffeine-induced increase in glucose uptake by the canine hindlimb.
The present study was designed to investigate the effects of alpha and beta adrenergic receptor blockers on caffeine-induced increase in canine hindlimb glucose uptake. The study was carried out on fasted male anaesthetized dogs divided into five groups [5 dogs per group]. Each dog was given a bolus injection of normal saline, caffeine [6 mg/kg] caffeine after pretreatment with prazosin [0.2 mg...
متن کاملGlucose uptake during centrally induced stress is insulin independent and enhanced by adrenergic blockade.
Glucose utilization increases markedly in the normal dog during stress induced by the intracerebroventricular (ICV) injection of carbachol. To determine the extent to which insulin, glucagon, and selective (alpha/beta)-adrenergic activation mediate the increment in glucose metabolic clearance rate (MCR) and glucose production (R(a)), we used five groups of normal mongrel dogs: 1) pancreatic cla...
متن کاملEffect of Curcumin, an Active Substance of Turmeric, on Acute Hyperglycemia Induced by Ketamine-Xylazine: Role of α2-Adrenergic Receptor
Objective- Medicinal plants and their active constituents are frequently used components for treating hyperglycemia. In the present study, the effect of curcumin was investigated on acute hyperglycemia and hyperinsulinemia induced by ketamine-xylazine in rats. To explore the possible mechanism, yohimbine (an α2-adrenergic receptor antagonist) was also used. Design...
متن کاملPossible Involvement of Glutamatergic, Adrenergic and Dopaminergic System in Methylphenidate - induced Motor Activity and Mood-related Alterations in Rats
Background and Objective: Methylphenidate (MPH), as a central nervous system stimulant, is often used to manage hyperactive disorders. The literature is scarce regarding the behavioral consequences of chronic MPH treatment and the role of involved receptors. Thus, in the current study involved receptors in MPH induced-anxiety, depression and motor activity disorders were evaluated. Materials a...
متن کاملThe glucose receptor. A defective mechanism in diabetes mellitus distinct from the beta adrenergic receptor.
Acute serum insulin responses in 10 normal subjects after rapid intravenous injection of glucose (5 g) or isoproterenol (2 mug) were of similar magnitude and timing (glucose: 431+/-349%; mean Delta3-5' insulin (IRI)+/-SD, per cent basal and isoproterenol: 359+/-216%; mean Delta2-4' IRI+/-SD, per cent basal). To elucidate the relationship of glucose-induced insulin secretion to pancreatic beta a...
